Effect of pH alterations on the vascular reactivity of dog isolated perfused mesenteric arteries.

چکیده
Accepted January 23, 1984 It is well recognized that the pH of the perfusing fluid is one of the important factors involved in the control of vascular reactivity. Respiratory acidosis diminishes the effect of vasoconstrictor substances on blood vessels (1). Rogers et al. (2) demonstrated that increasing pH potentiated the increase in perfusion pressure to administered catechol amines in isolated perfused mesenteric arteries. Recently, a new model of an isolated arterial preparation was developed by Hongo and Chiba (3) and modified by Tsuji and Chiba (4). By using this method, we have attempted to evaluate responses of dog isolated mesenteric arteries to several vasoconstrictor substances at low, normal and high pH levels of perfusing fliud. Twelve mongrel dogs weighing 9-16 kg were anesthetized with sodium pentobarbital (30 mg/kg, i.v.) and sacrificed by rapid exanguination, after treatment with sodium heparin (500 units/kg, i.v.). The mesenteric artery was dissected and carefully removed. Isolated mesenteric arteries selected for study were 10-15 mm in length and 0.5 1.0 mm in outer diameter, and they were cannulated as described previously (3, 4). Briefly, a steel cannula (21-27 gauge, 0.40-0.83 mm in outer diameter and 3 cm in length) was prepared with one end sealed. Near the sealed end of the cannula, 1 to 3 small holes were opened on the wall of the cannula. The steel cannula was inserted into each artery so that the small holes were covered with the proximal end of the artery, and this proximal end was fixed to the cannula by a thin thread. The isolated, cannulated artery was placed in a bath, which was maintained at constant temper ature of 37'C, and was perfused with

Relaxatory Effect of Gamma-Aminobutyric Acid (GABA) is Mediated by Same Pathway in Diabetic and Normal Rat Mesenteric Bed vessel
Objective(s) Diabetes related dysfunction of resistance vessels is associated with vascular occlusive diseases. Vasorelaxant agents may have a role in control of diabetic cardiovascular complications. Gamma aminobutyric acid (GABA) has demonstrated to cause vasorelaxation. The present study was designed to determine i) the vasorelaxatory effect of GABA on diabetic vessels and ii) the role of e...
متن کاملEvaluation of vascular effects of aqueous Crocus sativus petals’ extract in the hypertensive rats
Background and Aim: Crocus sativus (C. sativus) petals attenuates smooth muscle tension and blood pressure in control animals. However the antihypertensive effect and its mechanisms haven’t been recognized. This study investigates the antihypertensive effects of C. sativus petals’ aqueous extract in hypertensive rats and also responses of the rat isolated perfused mesenteric bed. Materials and...
متن کاملChanges in vascular reactivity of the coronary artery and thoracic aorta in the delta sarcoglycan null mutant mice
Introduction: Mutations in the delta sarcoglycan gene (d-SG) cause limb-girdle muscular dystrophy type 2F with structural and functional alterations in cardiac, smooth and skeletal muscle. The objective of the present study was to improve information about changes in vascular reactivity of the thoracic aorta and the coronary artery in the perfused heart of the d-SG-null mutant mouse model. ...
متن کاملEffects of estrogens and selective estrogen receptor modulators (SERMs) on vascular reactivity in the perfused mesenteric vascular bed Running head: Mesenteric vascular responses to estrogens and SERMs
Estrogens, and SERMs such as raloxifene (RAL) and tamoxifen (TAM), acutely relax arteries, but the long term effects of estrogens and SERMs on vascular reactivity in the mesenteric vasculature have not been well-defined. In this study, we used an isolated perfused mesenteric vascular bed technique to investigate the effect of chronic treatment of estrogens and SERMs on vascular reactivity of th...

1 Hypertensive and normotensive rats of the same age group were isolated from an inbred colony of spontaneously hypertensive rats. 2 The perfused hindquarter and mesenteric artery preparations obtained from hypertensive and normotensive rats exhibited an increased reactivity to noradrenaline (NA) and angiotensin II. 3 Dose-response curves to NA obtained from hypertensive and normotensive rats e...
